Century-old ocean data provides further confirmation of global warming

May 28, 2013 — A new NASA and university analysis of ocean data collected more than 135 years ago by the crew of the HMS Challenger oceanographic expedition provides further confirmation that human activities have warmed our planet over the past century.






Researchers from the University of Tasmania, Sandy Bay, Australia; and NASA's Jet Propulsion Laboratory, Pasadena, Calif., combined the ship's measurements of ocean temperatures with modern observations from the international Argo array of ocean profiling floats. They used both as inputs to state-of-the-art climate models, to get a picture of how the world's oceans have changed since the Challenger's voyage.

The Challenger expedition, from 1872 to 1876, was the world's first global scientific survey of life beneath the ocean surface. Along the way, scientists measured ocean temperatures, lowering thermometers hundreds of meters deep on ropes.

"The key to this research was to determine the range of uncertainty for the measurements taken by the crew of the Challenger," said Josh Willis, a JPL climate scientist and NASA project scientist for the upcoming U.S./European Jason-3 oceanography satellite, scheduled to launch in 2015. "After we had taken all these uncertainties into account, it became apparent that the rate of warming we saw across the oceans far exceeded the degree of uncertainty around the measurements. So, while the uncertainty was large, the warming signal detected was far greater."

Uncertainties around the Challenger's measurements were caused by the limited areas measured during the voyage; the actual depths the thermometers descended to; and the likely natural variation in temperature that could occur in each region during the voyage.

"Our research revealed warming of the planet can be clearly detected since 1873 and that our oceans continue to absorb the great majority of this heat," said researcher and lead author Will Hobbs of the University of Tasmania's Institute for Marine and Antarctic Studies and the Australian Research Council's Centre of Excellence for Climate System Science. "Currently, scientists estimate the oceans absorb more than 90 percent of the heat trapped by greenhouse gases, and we attribute the global warming to anthropogenic (human-produced) causes."

The Challenger expedition measurements also revealed that thermal expansion of sea water caused by global warming contributed about 40 percent of the total sea level rise seen in tide gauges from 1873 to 1955. The remaining 60 percent was likely to have come from the melting of ice sheets and glaciers. Prior to this research, climate models offered the only way to estimate the change before the 1950s.

Results of the study are published in the journal Geophysical Research Letters.

林登国:一位台湾收藏家的玉石情缘

  新华网太原5月27日电(记者王学涛)57岁的林登国拥有500多件古玉,他甚至一度为玉“疯狂”.然而,随着年岁渐长,现实的压力接踵而至.这位在台湾收藏了25年古玉的“好玉者”在去年做出了一个重要决定:第一次到大陆,开始为自己的千年玉石寻找有缘人.

  “一个月有三周在大陆,这种心情就如同嫁女儿,希望为它们找到好‘婆家’.”林登国说.

  为了收藏古玉,林登国在很多年前卖掉了家里祖传的土地和房子,和妻女长年租房子住.“每次看到没有研究过的玉,就想买下,研究古玉要有实物.”林登国说,为此,他几乎花掉了家里所有积蓄.如今,林登国已是台湾玉器交流协会的资深委员.

  “中华文化和玉器紧密相连.”林登国告诉记者,中华大地的先民曾认识到玉是“美而不朽”的石头,最开始被当作一种神器,后来在儒学中以玉比德,成为权力、身份的象征.

  在日前举行的2013年全国文物艺术品山西交流会上,林登国带着多件汉代以前的古玉前来寻找合适的买家.尽管卖玉并非他所愿.

  林登国说,自己年纪大了,体力和眼力都不如从前.此外,女儿也到了谈婚论嫁的年龄,而他现在还要为每月5000元的房租操心.迫于现实压力,他希望给这些玉器找到好归宿.

  虽然这次山西之行仅卖出3件古玉,但林登国仍感到欣慰.

  “从购买者的眼神就能看到其对玉石的内心感情.”林登国说,他不愿意卖给投资商.

  谈到500多件古玉的未来,林登国早已做好规划.他说,除了一部分要留在学校供教学外,剩余的要一件件为它们找到“有缘人”.

  “这些玉石有几千年的生命,而我只是它们生命中的过客.”林登国说,他正准备写一本书,将自己25年来学习、研究、鉴定古玉的心得体会整理出来,为传承中华文化贡献微薄之力.

Formula for turning cement into 'metal'

"This new material has lots of applications, including as thin-film resistors used in liquid-crystal displays, basically the flat panel computer monitor that you are probably reading this from at the moment," said Chris Benmore, a physicist from the U.S. Department of Energy's (DOE) Argonne National Laboratory who worked with a team of scientists from Japan, Finland and Germany to take the "magic" out of the cement-to-metal transformation. Benmore and Shinji Kohara from Japan Synchrotron Radiation Research Institute/SPring-8 led the research effort.

This change demonstrates a unique way to make metallic-glass material, which has positive attributes including better resistance to corrosion than traditional metal, less brittleness than traditional glass, conductivity, low energy loss in magnetic fields, and fluidity for ease of processing and molding. Previously, only metals have been able to transition to a metallic-glass form. Cement does this by a process called electron trapping, a phenomena only previously seen in ammonia solutions. Understanding how cement joined this exclusive club opens the possibility of turning other solid normally insulating materials into room-temperature semiconductors.

"This phenomenon of trapping electrons and turning liquid cement into liquid metal was found recently, but not explained in detail until now," Benmore said. "Now that we know the conditions needed to create trapped electrons in materials we can develop and test other materials to find out if we can make them conduct electricity in this way."

The results were reported May 27 in the journal the Proceeding of the National Academy of Sciences in the article "Network topology for the formation of solvated electrons in binary CaO-Al2O3 composition glasses."

The team of scientists studied mayenite, a component of alumina cement made of calcium and aluminum oxides. They melted it at temperatures of 2,000 degrees Celsius using an aerodynamic levitator with carbon dioxide laser beam heating. The material was processed in different atmospheres to control the way that oxygen bonds in the resulting glass. The levitator keeps the hot liquid from touching any container surfaces and forming crystals. This let the liquid cool into glassy state that can trap electrons in the way needed for electronic conduction. The levitation method was developed specifically for in-situ measurement at Argonne's Advanced Photon Source by a team led by Benmore.

The scientists discovered that the conductivity was created when the free electrons were "trapped" in the cage-like structures that form in the glass. The trapped of electrons provided a mechanism for conductivity similar to the mechanism that occurs in metals.

To uncover the details of this process, scientists combined several experimental techniques and analyzed them using a supercomputer. They confirmed the ideas in experiments using different X-ray techniques at Spring 8 in Japan combined with earlier measurements at the Intense Pulsed Neutron Source and the Advanced Photon Source.

Research was supported by the Ministry of Education, Culture, Sports, Science, and Technology of Japan, the Japan Science and Technology Agency, and the Academy of Finland.

Stem cell injections improve spinal injuries in rats

May 27, 2013 — An international team led by researchers at the University of California, San Diego School of Medicine reports that a single injection of human neural stem cells produced neuronal regeneration and improvement of function and mobility in rats impaired by an acute spinal cord injury (SCI).






The findings are published in the May 28, 2013 online issue of Stem Cell Research & Therapy.

Martin Marsala, MD, professor in the Department of Anesthesiology, with colleagues at UC San Diego and in Slovakia, the Czech Republic and The Netherlands, said grafting neural stem cells derived from a human fetal spinal cord to the rats' spinal injury site produced an array of therapeutic benefits -- from less muscle spasticity to new connections between the injected stem cells and surviving host neurons.

"The primary benefits were improvement in the positioning and control of paws during walking tests and suppression of muscle spasticity," said Marsala, a specialist in spinal cord trauma and spinal injury-related disorders. Spasticity -- exaggerated muscle tone or uncontrolled spasms -- is a serious and common complication of traumatic injury to the spinal cord.

The human stem cells, said the scientists, appeared to vigorously take root at the injury site.

"In all cell-grafted animals, there was robust engraftment, and neuronal maturation of grafted human neurons was noted," Marsala said. "Importantly, cysts or cavities that can form in or around spinal injuries were not present in any cell-treated animal. The injury-caused cavity was completely filled by grafted cells."

The rats received the pure stem cell grafts three days after injury (no other supporting materials were used) and were given drugs to suppress an immune response to the foreign stem cells. Marsala said grafting at any time after the injury appears likely to work in terms of blocking the formation of spinal injury cavities, but that more work would be required to determine how timing affects functional neurological benefit.

The grafted stem cells, according to Marsala, appear to be doing two things: stimulating host neuron regeneration and partially replacing the function of lost neurons.

"Grafted spinal stem cells are rich source of different growth factors which can have a neuroprotective effect and can promote sprouting of nerve fibers of the host neurons. We have also demonstrated that grafted neurons can develop contacts with the host neurons and, to some extent, restore the connectivity between centers, above and below the injury, which are involved in motor and sensory processing."

The scientists used a line of human embryonic stem cells recently approved for Phase 1 human trials in patients with chronic traumatic spinal injuries. Marsala said the ultimate goal is to develop neural precursor cells (capable of becoming any of the three main cell types in the nervous system) from induced pluripotent stem cells derived from patients, which would likely eliminate the need for immunosuppression treatment.

Pending approval by UC San Diego's Institutional Review Board, the next step is a small phase 1 trial to test safety and efficacy with patients who have suffered a thoracic spinal cord injury (between vertebrae T2-T12) one to two years earlier, and who have no motor or sensory function at or below the spinal injury site.

"This is exciting, especially because, historically, there has been very little to offer patients with acute spinal cord injury," said study co-author Joseph Ciacci, MD, professor of surgery and program director of the Neurosurgery Residency at the UC San Diego School of Medicine. Ciacci, who is also chief of neurosurgery for the Veterans Affairs San Diego Healthcare System, will oversee the clinical trial at UC San Diego and the VA.

Ciacci said if the initial study confirms safety and efficacy, as well as the viability of the implanted cells, neural regeneration and decreased spasticity, the protocol can be expanded to other patients with other forms of severe spinal cord injury.

Formula for turning cement into 'metal'

"This new material has lots of applications, including as thin-film resistors used in liquid-crystal displays, basically the flat panel computer monitor that you are probably reading this from at the moment," said Chris Benmore, a physicist from the U.S. Department of Energy's (DOE) Argonne National Laboratory who worked with a team of scientists from Japan, Finland and Germany to take the "magic" out of the cement-to-metal transformation. Benmore and Shinji Kohara from Japan Synchrotron Radiation Research Institute/SPring-8 led the research effort.

This change demonstrates a unique way to make metallic-glass material, which has positive attributes including better resistance to corrosion than traditional metal, less brittleness than traditional glass, conductivity, low energy loss in magnetic fields, and fluidity for ease of processing and molding. Previously, only metals have been able to transition to a metallic-glass form. Cement does this by a process called electron trapping, a phenomena only previously seen in ammonia solutions. Understanding how cement joined this exclusive club opens the possibility of turning other solid normally insulating materials into room-temperature semiconductors.

"This phenomenon of trapping electrons and turning liquid cement into liquid metal was found recently, but not explained in detail until now," Benmore said. "Now that we know the conditions needed to create trapped electrons in materials we can develop and test other materials to find out if we can make them conduct electricity in this way."

The results were reported May 27 in the journal the Proceeding of the National Academy of Sciences in the article "Network topology for the formation of solvated electrons in binary CaO-Al2O3 composition glasses."

The team of scientists studied mayenite, a component of alumina cement made of calcium and aluminum oxides. They melted it at temperatures of 2,000 degrees Celsius using an aerodynamic levitator with carbon dioxide laser beam heating. The material was processed in different atmospheres to control the way that oxygen bonds in the resulting glass. The levitator keeps the hot liquid from touching any container surfaces and forming crystals. This let the liquid cool into glassy state that can trap electrons in the way needed for electronic conduction. The levitation method was developed specifically for in-situ measurement at Argonne's Advanced Photon Source by a team led by Benmore.

The scientists discovered that the conductivity was created when the free electrons were "trapped" in the cage-like structures that form in the glass. The trapped of electrons provided a mechanism for conductivity similar to the mechanism that occurs in metals.

To uncover the details of this process, scientists combined several experimental techniques and analyzed them using a supercomputer. They confirmed the ideas in experiments using different X-ray techniques at Spring 8 in Japan combined with earlier measurements at the Intense Pulsed Neutron Source and the Advanced Photon Source.

Research was supported by the Ministry of Education, Culture, Sports, Science, and Technology of Japan, the Japan Science and Technology Agency, and the Academy of Finland.

Stem cell injections improve spinal injuries in rats

May 27, 2013 — An international team led by researchers at the University of California, San Diego School of Medicine reports that a single injection of human neural stem cells produced neuronal regeneration and improvement of function and mobility in rats impaired by an acute spinal cord injury (SCI).






The findings are published in the May 28, 2013 online issue of Stem Cell Research & Therapy.

Martin Marsala, MD, professor in the Department of Anesthesiology, with colleagues at UC San Diego and in Slovakia, the Czech Republic and The Netherlands, said grafting neural stem cells derived from a human fetal spinal cord to the rats' spinal injury site produced an array of therapeutic benefits -- from less muscle spasticity to new connections between the injected stem cells and surviving host neurons.

"The primary benefits were improvement in the positioning and control of paws during walking tests and suppression of muscle spasticity," said Marsala, a specialist in spinal cord trauma and spinal injury-related disorders. Spasticity -- exaggerated muscle tone or uncontrolled spasms -- is a serious and common complication of traumatic injury to the spinal cord.

The human stem cells, said the scientists, appeared to vigorously take root at the injury site.

"In all cell-grafted animals, there was robust engraftment, and neuronal maturation of grafted human neurons was noted," Marsala said. "Importantly, cysts or cavities that can form in or around spinal injuries were not present in any cell-treated animal. The injury-caused cavity was completely filled by grafted cells."

The rats received the pure stem cell grafts three days after injury (no other supporting materials were used) and were given drugs to suppress an immune response to the foreign stem cells. Marsala said grafting at any time after the injury appears likely to work in terms of blocking the formation of spinal injury cavities, but that more work would be required to determine how timing affects functional neurological benefit.

The grafted stem cells, according to Marsala, appear to be doing two things: stimulating host neuron regeneration and partially replacing the function of lost neurons.

"Grafted spinal stem cells are rich source of different growth factors which can have a neuroprotective effect and can promote sprouting of nerve fibers of the host neurons. We have also demonstrated that grafted neurons can develop contacts with the host neurons and, to some extent, restore the connectivity between centers, above and below the injury, which are involved in motor and sensory processing."

The scientists used a line of human embryonic stem cells recently approved for Phase 1 human trials in patients with chronic traumatic spinal injuries. Marsala said the ultimate goal is to develop neural precursor cells (capable of becoming any of the three main cell types in the nervous system) from induced pluripotent stem cells derived from patients, which would likely eliminate the need for immunosuppression treatment.

Pending approval by UC San Diego's Institutional Review Board, the next step is a small phase 1 trial to test safety and efficacy with patients who have suffered a thoracic spinal cord injury (between vertebrae T2-T12) one to two years earlier, and who have no motor or sensory function at or below the spinal injury site.

"This is exciting, especially because, historically, there has been very little to offer patients with acute spinal cord injury," said study co-author Joseph Ciacci, MD, professor of surgery and program director of the Neurosurgery Residency at the UC San Diego School of Medicine. Ciacci, who is also chief of neurosurgery for the Veterans Affairs San Diego Healthcare System, will oversee the clinical trial at UC San Diego and the VA.

Ciacci said if the initial study confirms safety and efficacy, as well as the viability of the implanted cells, neural regeneration and decreased spasticity, the protocol can be expanded to other patients with other forms of severe spinal cord injury.

Scientists find mechanism that causes noise-induced tinnitus and drug that can prevent it

May 27, 2013 — An epilepsy drug shows promise in an animal model at preventing tinnitus from developing after exposure to loud noise, according to a new study by researchers at the University of Pittsburgh School of Medicine. The findings, reported this week in the early online version of the Proceedings of the National Academy of Sciences, reveal for the first time the reason the chronic and sometimes debilitating condition occurs.






An estimated 5 to 15 percent of Americans hear whistling, clicking, roaring and other phantom sounds of tinnitus, which typically is induced by exposure to very loud noise, said senior investigator Thanos Tzounopoulos, Ph.D., associate professor and member of the auditory research group in the Department of Otolaryngology, Pitt School of Medicine.

"There is no cure for it, and current therapies such as hearing aids don't provide relief for many patients," he said. "We hope that by identifying the underlying cause, we can develop effective interventions."

The team focused on an area of the brain that is home to an important auditory center called the dorsal cochlear nucleus (DCN). From previous research in a mouse model, they knew that tinnitus is associated with hyperactivity of DCN cells -- they fire impulses even when there is no actual sound to perceive. For the new experiments, they took a close look at the biophysical properties of tiny channels, called KCNQ channels, through which potassium ions travel in and out of the cell.

"We found that mice with tinnitus have hyperactive DCN cells because of a reduction in KCNQ potassium channel activity," Dr. Tzounopoulos said. "These KCNQ channels act as effective "brakes" that reduce excitability or activity of neuronal cells."

In the model, sedated mice are exposed in one ear to a 116-decibel sound, about the loudness of an ambulance siren, for 45 minutes, which was shown in previous work to lead to the development of tinnitus in 50 percent of exposed mice. Dr. Tzounopoulos and his team tested whether an FDA-approved epilepsy drug called retigabine, which specifically enhances KCNQ channel activity, could prevent the development of tinnitus. Thirty minutes into the noise exposure and twice daily for the next five days, half of the exposed group was given injections of retigabine.

Seven days after noise exposure, the team determined whether the mice had developed tinnitus by conducting startle experiments, in which a continuous, 70 dB tone is played for a period, then stopped briefly and then resumed before being interrupted with a much louder pulse. Mice with normal hearing perceive the gap in sounds and are aware something had changed, so they are less startled by the loud pulse than mice with tinnitus, which hear phantom noise that masks the moment of silence in between the background tones.

The researchers found that mice that were treated with retigabine immediately after noise exposure did not develop tinnitus. Consistent with previous studies, 50 percent of noise-exposed mice that were not treated with the drug exhibited behavioral signs of the condition.

"This is an important finding that links the biophysical properties of a potassium channel with the perception of a phantom sound," Dr. Tzounopoulos said. "Tinnitus is a channelopathy, and these KCNQ channels represent a novel target for developing drugs that block the induction of tinnitus in humans."

The KCNQ family is composed of five different subunits, four of which are sensitive to retigabine. He and his collaborators aim to develop a drug that is specific for the two KCNQ subunits involved in tinnitus to minimize the potential for side effects.

"Such a medication could be a very helpful preventive strategy for soldiers and other people who work in situations where exposure to very loud noise is likely," Dr. Tzounopoulos said. "It might also be useful for other conditions of phantom perceptions, such as pain in a limb that has been amputated."

Like many mammals, rats have their eyes on the sides of their heads. This gives them a very wide visual field, useful for detection of predators. However, three-dimensional vision requires overlap of the visual fields of the two eyes. Thus, the visual system of these animals needs to meet two conflicting demands at the same time; on the one hand maximum surveillance and on the other hand detailed binocular vision.

The research team from the Max Planck Institute for Biological Cybernetics have now, for the first time, observed and characterised the eye movements of freely moving rats. They fitted minuscule cameras weighing only about one gram to the animals' heads, which could record the lightning-fast eye movements with great precision. The scientists also used another new method to measure the position and direction of the head, enabling them to reconstruct the rats' exact line of view at any given time.

The Max Planck scientists' findings came as a complete surprise. Although rats process visual information from their eyes through very similar brain pathways to other mammals, their eyes evidently move in a totally different way. "Humans move their eyes in a very stereotypical way for both counteracting head movements and searching around. Both our eyes move together and always follow the same object. In rats, on the other hand, the eyes generally move in opposite directions," explains Jason Kerr from the Max Planck Institute for Biological Cybernetics.

In a series of behavioural experiments, the neurobiologists also discovered that the eye movements largely depend on the position of the animal's head. "When the head points downward, the eyes move back, away from the tip of the nose. When the rat lifts its head, the eyes look forward: cross-eyed, so to speak. If the animal puts its head on one side, the eye on the lower side moves up and the other eye moves down." says Jason Kerr.

In humans, the direction in which the eyes look must be precisely aligned, otherwise an object cannot be fixated. A deviation measuring less than a single degree of the field of view is enough to cause double vision. In rats, the opposing eye movements between left and right eye mean that the line of vision varies by as much as 40 degrees in the horizontal plane and up to 60 degrees in the vertical plane. The consequence of these unusual eye movements is that irrespective of vigorous head movements in all planes, the eyes movements always move in such a way to ensure that the area above the animal is always in view simultaneously by both eyes -something that does not occur in any other region of the rat's visual field.

These unusual eye movements that rats possess appear to be the visual system's way of adapting to the animals' living conditions, given that they are preyed upon by numerous species of birds. Although the observed eye movements prevent the fusion of the two visual fields, the scientists postulate that permanent visibility in the direction of potential airborne attackers dramatically increases the animals' chances of survival.

Dr. Tzounopoulos and his team tested whether an FDA-approved epilepsy drug called retigabine, which specifically enhances KCNQ channel activity, could prevent the development of tinnitus. Thirty minutes into the noise exposure and twice daily for the next five days, half of the exposed group was given injections of retigabine.

Seven days after noise exposure, the team determined whether the mice had developed tinnitus by conducting startle experiments, in which a continuous, 70 dB tone is played for a period, then stopped briefly and then resumed before being interrupted with a much louder pulse. Mice with normal hearing perceive the gap in sounds and are aware something had changed, so they are less startled by the loud pulse than mice with tinnitus, which hear phantom noise that masks the moment of silence in between the background tones.

The researchers found that mice that were treated with retigabine immediately after noise exposure did not develop tinnitus. Consistent with previous studies, 50 percent of noise-exposed mice that were not treated with the drug exhibited behavioral signs of the condition.

"This is an important finding that links the biophysical properties of a potassium channel with the perception of a phantom sound," Dr. Tzounopoulos said. "Tinnitus is a channelopathy, and these KCNQ channels represent a novel target for developing drugs that block the induction of tinnitus in humans."

The KCNQ family is composed of five different subunits, four of which are sensitive to retigabine. He and his collaborators aim to develop a drug that is specific for the two KCNQ subunits involved in tinnitus to minimize the potential for side effects.

"Such a medication could be a very helpful preventive strategy for soldiers and other people who work in situations where exposure to very loud noise is likely," Dr. Tzounopoulos said. "It might also be useful for other conditions of phantom perceptions, such as pain in a limb that has been amputated."

Advanced biological computer developed

May 23, 2013 — Using only biomolecules (such as DNA and enzymes), scientists at the Technion-Israel Institute of Technology have developed and constructed an advanced biological transducer, a computing machine capable of manipulating genetic codes, and using the output as new input for subsequent computations. The breakthrough might someday create new possibilities in biotechnology, including individual gene therapy and cloning.






The findings appear today (May 23, 2013) in Chemistry & Biology (Cell Press).

Interest in such biomolecular computing devices is strong, mainly because of their ability (unlike electronic computers) to interact directly with biological systems and even living organisms. No interface is required since all components of molecular computers, including hardware, software, input and output, are molecules that interact in solution along a cascade of programmable chemical events.

"Our results show a novel, synthetic designed computing machine that computes iteratively and produces biologically relevant results," says lead researcher Prof. Ehud Keinan of the Technion Schulich Faculty of Chemistry. "In addition to enhanced computation power, this DNA-based transducer offers multiple benefits, including the ability to read and transform genetic information, miniaturization to the molecular scale, and the aptitude to produce computational results that interact directly with living organisms."

The transducer could be used on genetic material to evaluate and detect specific sequences, and to alter and algorithmically process genetic code. Similar devices, says Prof. Keinan, could be applied for other computational problems.

"All biological systems, and even entire living organisms, are natural molecular computers. Every one of us is a biomolecular computer, that is, a machine in which all components are molecules "talking" to one another in a logical manner. The hardware and software are complex biological molecules that activate one another to carry out some predetermined chemical tasks. The input is a molecule that undergoes specific, programmed changes, following a specific set of rules (software) and the output of this chemical computation process is another well defined molecule."

Also contributing to the research were postdoctoral fellows Dr. Tamar Ratner and Dr. Ron Piran of the Technion's Schulich Faculty of Chemistry, and Dr. Natasha Jonoska of the Department of Mathematics at the University of South Florida.
